Biological Background: DGKA Function and Localization

  • DGKA (Diacylglycerol kinase alpha), also known as 80 kDa diacylglycerol kinase or DGK-alpha, is a lipid kinase that phosphorylates diacylglycerol (DAG) to produce phosphatidic acid (PA), thereby acting as a critical switch between DAG- and PA-mediated signaling pathways. Related references: PMID:15544348, PMID:2175712
  • It also phosphorylates 1-alkyl-2-acylglycerol in vitro, with a preference for substrates containing an arachidonoyl group, and participates in the biosynthesis of complex lipids. Related references: PMID:15544348
  • DGKA is involved in the production of alkyl-lysophosphatidic acid through phosphorylation of 1-alkyl-2-acetyl glycerol. Related references: PMID:22627129
  • The protein contains zinc-finger domains and an ATP-binding site, classifying it as a transferase and a calcium-sensitive enzyme.
  • DGKA is primarily localized in the cytoplasm and cytosol.
  • It is expressed in lymphocytes, suggesting a role in immune cell signaling.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-200), so the antibody may detect full-length DGKA and N-terminal fragments. For WB, expect a band at ~83 kDa; consider using knockdown or knockout controls to confirm specificity.
  • For IHC-P, optimize antigen retrieval conditions and antibody dilution; lymphoid tissues from human, mouse, or rat are suitable positive controls.
  • For ELISA, use recombinant DGKA protein as a standard and assess cross-reactivity with other DGK isoforms if needed.
