Biological Background: Diacylglycerol kinase alpha Function and Localization
- Diacylglycerol kinase alpha (DGKA), also known as 80 kDa diacylglycerol kinase or DGK-alpha, is a lipid kinase that phosphorylates diacylglycerol (DAG) to generate phosphatidic acid (PA).
- By interconverting DAG and PA, DGKA serves as a central switch between signaling pathways involving these two second messengers, which have contrasting cellular effects. Related references: PMID:15544348, PMID:2175712
- DGKA can also phosphorylate 1-alkyl-2-acylglycerol (if it contains an arachidonoyl group) and 1-alkyl-2-acetyl glycerol, contributing to the production of alkyl-lysophosphatidic acid, another bioactive lipid. Related references: PMID:15544348, PMID:22627129
- Plays a role in the biosynthesis of complex lipids (Probable).
- Localizes primarily to the cytoplasm and cytosol.
- Predominantly expressed in lymphocytes, suggesting a role in immune cell signaling.
- Contains conserved domains including zinc-finger motifs, an ATP-binding site, and a calcium-binding region; subject to post-translational acetylation and alternative splicing.
- Theoretical molecular weight is approximately 82.6 kDa.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 1–200) of human DGKA; consider that epitopes within conserved domains may lead to cross-reactivity with other DGK family members.
- For Western blot, a band at ~83 kDa is expected; validate with appropriate positive controls such as lymphocyte lysates or DGKA-overexpressing cell lines.
- In IHC-P, optimize antigen retrieval and antibody dilution, as epitopes may be masked by fixation.
- ELISA performance may depend on plate coating and detection reagents; pairing with a suitable secondary antibody is advised.
