Biological Background: DGAT1 Function and Localization
- DGAT1 (Diacylglycerol O-acyltransferase 1), also known as ARAT, DGAT, ARGP1, DIAR7, catalyzes the terminal and only committed step in triacylglycerol synthesis using diacylglycerol and fatty acyl CoA as substrates. Related references: PMID:16214399, PMID:18768481, PMID:28420705
- In the small intestine, DGAT1 is highly expressed in epithelial cells and is essential for the absorption of dietary fats. Related references: PMID:18768481
- In the liver, DGAT1 plays a role in esterifying exogenous fatty acids to glycerol and is required for fat storage. Related references: PMID:16214399
- DGAT1 is present in female mammary glands, where it produces fat in the milk (By similarity).
- The enzyme may be involved in very low density lipoprotein (VLDL) assembly. Related references: PMID:18768481
- As the major acyl-CoA retinol acyltransferase (ARAT) in the skin, DGAT1 maintains retinoid homeostasis and prevents retinoid toxicity. Related references: PMID:16214399
- DGAT1 also exhibits additional acyltransferase activities, including monoacylglycerol acyltransferase (MGAT) and wax synthase activities, and can use 1-monoalkylglycerol as an acyl acceptor. Related references: PMID:28420705
- Subcellular localization: DGAT1 is an endoplasmic reticulum membrane protein.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 201-300 of human DGAT1 (NP_036211.2). This region may be conserved across species, supporting the observed cross-reactivity with mouse and rat.
- For Western blotting, the predicted molecular weight of DGAT1 is ~55 kDa. Validate the antibody with appropriate positive and negative controls in your sample system.
- In immunofluorescence and ELISA applications, consider optimizing the dilution factor and blocking conditions based on the specific sample type and experimental design.
- Cross-reactivity with human, mouse, and rat suggests potential utility in rodent models; however, independent validation in the relevant tissue or cell line is recommended.
