Biological Background: Desmoglein-3 Function and Localization
- Identified as the 130 kDa pemphigus vulgaris antigen (PVA), DSG3 is a member of the cadherin family. It is predominantly expressed in the basal and spinous layers of the epidermis, the outer root sheath of hair follicles, and mucosal epithelia (esophagus, oral cavity, tongue, tonsil) (PubMed:19717567, PubMed:9701552, PubMed:24220297, PubMed:16740002). Related references: PMID:19717567, PMID:9701552, PMID:24220297, PMID:16740002
- DSG3 is a core component of desmosomal junctions and is required for Ca²⁺-dependent cell-cell adhesion, particularly in keratinocytes subjected to mechanical stress (PubMed:31835537). Related references: PMID:31835537
- It is essential for anchoring the telogen hair club to the follicular outer root sheath, thereby securing the hair shaft during the resting phase (PubMed:9701552). Related references: PMID:9701552
- DSG3 maintains epithelial barrier integrity by promoting desmosome-mediated attachment of suprabasal to basal cells and by stabilizing plaque proteins DSP, JUP, PKP1, PKP2, and PKP3 (PubMed:22294297). Related references: PMID:22294297
- The protein participates in mechanotransduction by facilitating the plasma membrane localization of YAP1 in response to mechanical strain, involving a complex with PKP1 and YWHAG, and may thereby influence YAP1-driven transcription and proliferation (PubMed:31835537). It also organizes cortical F-actin and anchors actin to tight junctions via RAC1 to regulate contractility (PubMed:22796473). Related references: PMID:31835537, PMID:22796473
- In the cytoplasm, DSG3 supports the localization of E-cadherin (CDH1) and β-catenin (CTNNB1) at developing adherens junctions, potentially through modulating SRC activity (PubMed:22294297). Moreover, it suppresses keratinocyte migration via p38MAPK inhibition, suggesting a role in wound healing modulation (PubMed:26763450). Related references: PMID:22294297, PMID:26763450
- Subcellularly, DSG3 localizes to the cell membrane, desmosomes, tight junctions, and cytoplasm. It is a transmembrane glycoprotein with extracellular cadherin repeats, calcium-binding properties, and a predicted molecular weight of approximately 107.5 kDa.
Experimental Guidance and Technical Tips
- The immunogen (a.a. 50–615) covers most of the extracellular domain and may allow detection of full-length DSG3 in both denaturing (WB) and native (IHC-P) conditions.
- For WB, a band at ~108 kDa is expected; confirm specificity using DSG3‑positive cell lines (e.g., A‑431, HaCaT) or tissue lysates from skin or mucosa.
- In IHC‑P, antigen retrieval may be required; anticipate staining at cell membranes and desmosomes in stratified epithelia such as skin, esophagus, or tonsil.
- For ELISA, optimization of coating and detection conditions is recommended; the antibody can be tested as a capture or detection reagent.
- Cross‑reactivity is confirmed for human; reactivity with other species has not been determined and should be validated in the relevant experimental system.
- As DSG3 is a major autoantigen in pemphigus vulgaris, this antibody may assist in diagnostic assay development or autoimmune disease research models.
