Biological Background: Desmin Function and Localization
- Desmin is a muscle-specific type III intermediate filament protein encoded by the DES gene, crucial for maintaining sarcomere structure and myofibril integrity.
- It interconnects Z-disks and myofibrils, linking them to the sarcolemmal cytoskeleton, nuclei, and mitochondria, providing mechanical strength during muscle contraction. Related references: PMID:25358400

- For Western blotting, a band at ~54 kDa is expected in human, mouse, or rat muscle tissue lysates; validate detection of post-translational modifications if relevant.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF-P), use frozen or paraffin-embedded muscle sections; antigen retrieval may be considered for formalin-fixed tissues.
