Biological Background: Dermcidin Function and Localization
- Dermcidin (DCD), also known as preproteolysin, is a secreted antimicrobial protein encoded by the DCD gene (AIDD, DSEP). It undergoes alternative splicing to generate multiple isoforms.
- It is found in sweat and exhibits potent antimicrobial activity against a range of pathogens, including E. coli, E. faecalis, S. aureus, and C. albicans, with optimal activity at sweat-like pH and salt conditions (PubMed:11694882, PubMed:23426625). Related references: PMID:11694882 PMID:23426625
- Dermcidin assembles into homohexameric complexes that insert into pathogen membranes, forming anion channels that disrupt the transmembrane potential and lead to microbial death (PubMed:23426625). Related references: PMID:23426625
- Beyond antimicrobial function, DCD exhibits proteolytic activity, cleaving peptide bonds on the C-terminal side of arginine and, to a lesser extent, lysine residues (PubMed:17448443). Related references: PMID:17448443
- It also promotes neuron survival and possesses phosphatase activity, and may bind IgG, suggesting roles in neural protection and immune modulation (PubMed:9736629). Related references: PMID:9736629
- Subcellularly, DCD is secreted into sweat and other biological fluids; it can also associate with membranes. It has been detected in urine at the protein level.
- Constitutively expressed in eccrine sweat gland cells, with secretion into sweat at concentrations of 1–10 µg/ml.
- Key post-translational modifications include acetylation and glycosylation (proteoglycan), and it binds metal ions such as zinc and manganese, which are relevant for its structure and function.
Experimental Guidance and Technical Tips
- The immunogen was a recombinant protein covering amino acids 20-110 of human DCD (NP_444513.1). For WB, consider testing a range of primary antibody dilutions (e.g., 1:500–1:2000) and validate in your specific sample matrix.
- As DCD is a secreted protein, optimal detection in WB may require analysis of conditioned media or urea-containing extracts. For tissue samples, ensure thorough homogenization to release the soluble 11kDa form.
- For ELISA, direct or sandwich formats may be evaluated; pairing with a matched detection antibody could improve assay sensitivity. Validate in relevant biological fluids (e.g., sweat, urine) given cross-reactivity with human and mouse orthologs.
- Cross-reactivity with mouse is reported, but always confirm species-specific detection in your experimental system using appropriate controls.
