Biological Background: DENND4A Function and Localization
- DENND4A, also known as C-myc promoter-binding protein (MYCPBP) and IRLB, is a DENN domain-containing protein that functions as a guanine nucleotide exchange factor (GEF) and a transcription regulator.
- It may activate RAB10 by promoting the exchange of GDP for GTP, thereby converting inactive GDP-bound Rab proteins to their active GTP-bound state.
- It binds to the ISRE-like element (interferon-stimulated response element) of the MYC P2 promoter, potentially regulating MYC expression. Related references: PMID:8056341
- Subcellular localization is primarily in the nucleus, consistent with its role in transcriptional regulation.
- Ubiquitously expressed, with highest levels in bone marrow, intermediate in peripheral blood lymphocytes, and lowest in spleen. In brain, breast, and prostate tissues, expression is higher in normal cells than in tumor cells.
- Expression is regulated in a growth- and cell cycle-dependent manner, suggesting involvement in proliferation control.
- The protein undergoes alternative splicing and phosphorylation, adding layers of functional regulation.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight of DENND4A is approximately 209 kDa. Consider using a low-percentage SDS-PAGE gel (e.g., 6-8%) and sufficient transfer time to ensure efficient transfer of high-molecular-weight proteins.
- As a polyclonal antibody, lot-to-lot consistency should be validated. Include appropriate positive and negative controls in your experiments to confirm specificity.
- The immunogen corresponds to amino acids 1500-1650 of human DENND4A (NP_005839.3). For experiments in mouse samples, verify sequence homology in the target region.
- For ELISA applications, optimal coating concentrations and detection conditions should be empirically determined.
- Given the nuclear localization of DENND4A, consider using nuclear-enriched lysates for improved detection in WB.
