Biological Background: Delta-sarcoglycan (SGCD) Function and Localization

  • Delta-sarcoglycan (SGCD) is also known as 35 kDa dystrophin-associated glycoprotein (35DAG) and is a component of the sarcoglycan complex.
  • The sarcoglycan complex is a subcomplex of the larger dystrophin-glycoprotein complex, which mechanically links the F-actin cytoskeleton to the extracellular matrix, primarily in muscle cells.
  • SGCD is most strongly expressed in skeletal and cardiac muscle, with lower expression in smooth muscle, and weak expression in brain and lung.
  • Subcellularly, delta-sarcoglycan localizes to the cell membrane, specifically the sarcolemma in muscle cells, and is associated with the cytoskeleton.
  • The protein is a transmembrane glycoprotein with a signal-anchor and contains disulfide bonds; it undergoes alternative splicing.
  • Mutations in SGCD are associated with cardiomyopathy and limb-girdle muscular dystrophy (LGMD), classifying it as a disease variant protein.

Experimental Guidance and Technical Tips

  • The immunogen targets amino acids 61-290 of human delta-sarcoglycan, a region that may include extracellular or intracellular domains; consider this when designing experimental protocols.
  • For Western blot (WB), the predicted molecular weight is approximately 32kDa; include appropriate positive controls such as skeletal or cardiac muscle lysates from human, mouse, or rat.
  • In immunofluorescence/immunocytochemistry (IF/ICC), validate the antibody’s localization to the sarcolemma using muscle tissue sections or appropriate cell lines.
  • For ELISA, optimize coating concentration and detection conditions empirically, as no standardized dilution is provided.
  • Given cross-reactivity with mouse and rat, this antibody can potentially be used in comparative studies; however, always validate specificity in your own sample system.
