Biological Background: DEFB4A (Beta-defensin 2) Function and Localization
- DEFB4A, also known as beta-defensin 2 (BD-2, hBD-2), SAP1, DEFB2, DEFB4, DEFB102, is a member of the beta-defensin family of antimicrobial peptides.

Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to amino acids 1-64 of human DEFB4A, which covers the N-terminal region including the signal peptide. Consider that the antibody may recognize the precursor and/or mature forms; validation in relevant biological systems is recommended.
- For Western blot (WB), due to the small molecular weight of ~7 kDa, use a high-percentage polyacrylamide gel (e.g., 15% or gradient gel) and carefully optimize transfer conditions to avoid over-transfer. Positive controls can include lysates from human lung or tracheal epithelial cells.
- In immunofluorescence on paraffin-embedded sections (IF-P), anticipate extracellular or vesicular staining patterns, as DEFB4A is a secreted protein. Permeabilization may be adjusted; include appropriate controls.
- For ELISA, the antibody can be used for detection of native or recombinant DEFB4A in biological fluids such as bronchoalveolar lavage or cell culture supernatants. Pairing with a capture antibody may be explored for sandwich ELISA development.
- This antibody is reactive with human DEFB4A; cross-reactivity with other species or defensin family members has not been tested as per available data. Validate specificity in your sample system.
