Biological Background: DEFA5 Function and Localization
- DEFA5 (Defensin alpha 5, HD5) is a host-defense peptide that maintains sterility in the urogenital system. Related references: PMID:12021776, PMID:12660734, PMID:15616305
- It exhibits antimicrobial activity against a broad spectrum of bacteria, including Gram-negative E. coli, P. aeruginosa, S. typhimurium, and Gram-positive S. aureus, B. cereus, L. monocytogenes, among others. Related references: PMID:12021776, PMID:15616305, PMID:19589339
- DEFA5 confers resistance to intestinal infection by S. typhimurium and also shows activity against enteric commensal bacteria such as B. adolescentis and L. acidophilus. Related references: PMID:12660734, PMID:25354318
- The peptide binds to bacterial membranes, causing membrane disintegration, and neutralizes B. anthracis lethal factor enzymatic activity. Related references: PMID:25782105, PMID:22573326
- It induces secretion of the chemokine IL-8 by intestinal epithelial cells. Related references: PMID:19589339
- As a defensin, DEFA5 is a secreted protein found in cytoplasmic vesicles and secretory vesicles (UniProt Q01523).
- Tissue expression is prominent in Paneth cells of the small intestine, urothelium, kidney collecting tubules, and female genital tract epithelia; endometrial expression varies with the menstrual cycle. Related references: PMID:12021776, PMID:1429669, PMID:22359618, PMID:9588893
- The mature peptide is a glycoprotein with multiple disulfide bonds, belonging to the defensin family, and functions in innate immunity as an antibiotic and antimicrobial agent (Keywords: Antibiotic, Antimicrobial, Defensin, Disulfide bond, Glycoprotein).
Experimental Guidance and Technical Tips
- For Western blotting, consider using high-percentage SDS-PAGE gels and low molecular weight markers due to the small ~10kDa size of DEFA5.
- Validate antibody specificity in lysates from DEFA5-expressing tissues such as intestinal Paneth cells, urogenital epithelia, or relevant cell lines.
- For ELISA applications, the antibody can be used as a capture or detection reagent; optimize pairing with a matched antibody pair for sandwich ELISA development.
- Since the immunogen corresponds to the mature peptide region (aa 20-94), the antibody is expected to recognize processed and secreted forms of DEFA5.
