Biological Background: Decorin Function and Localization
- Decorin, also known as bone proteoglycan II or PG40, is a secreted proteoglycan encoded by the DCN gene (SLRR1B) in humans. It belongs to the small leucine-rich proteoglycan (SLRP) family.
- The protein features multiple leucine-rich repeats, which mediate protein-protein interactions and contribute to its role in the extracellular matrix.
- Decorin's primary function is the regulation of collagen fibrillogenesis; it may affect the rate of fibril formation.
- It is secreted into the extracellular space and is a component of the extracellular matrix. Tissue expression has been detected in placenta, cerebrospinal fluid, fibroblasts, and urine. Related references: PMID:32337544 PMID:25326458 PMID:36213313 PMID:37453717
- Post-translational modifications include glycosylation and disulfide bond formation. Alternative splicing has also been noted.
- Keywords associated with decorin include: Secreted, extracellular matrix, glycoprotein, leucine-rich repeat, proteoglycan, disulfide bond, and corneal dystrophy (mutations in DCN are linked to congenital stromal corneal dystrophy).
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 31-359 of human decorin (mature protein after signal peptide cleavage), so the antibody may recognize both glycosylated and non-glycosylated forms.
- For Western blotting, a band at approximately 40 kDa is expected; however, post-translational modifications such as glycosylation can cause a shift in molecular weight.
- Polyclonal antibodies can exhibit lot-to-lot variability; consider titrating and validating the antibody in your specific sample type and application.
- Indicated cross-reactivity with mouse and rat; verify reactivity in your species of interest if critical.
- For IHC-P, optimization of antigen retrieval and antibody dilution is recommended.
- In ELISA, pairing with a suitable capture/detection antibody may enhance performance.
