Biological Background: DDX5 Function and Localization

  • DDX5, also known as p68, is a member of the DEAD box protein family and functions as a probable ATP-dependent RNA helicase.
  • It plays a role in alternative pre-mRNA splicing, such as increasing tau exon 10 inclusion in an RBM4-dependent manner, and its helicase activity is stimulated by single-stranded RNA.
  • DDX5 acts as a transcriptional coregulator: it coactivates the androgen receptor (AR), MYOD1 (involved in skeletal muscle differentiation), TP53 (in response to DNA damage), and RUNX2 (osteoblast differentiation), and can also repress transcription via association with histone deacetylases.
  • As part of a large PER complex, DDX5 is involved in circadian rhythm regulation by inhibiting 3' transcriptional termination of clock genes like PER1 and NR1D1.
  • The protein localizes predominantly to the nucleus, including nucleoli and nuclear speckles, but can also be found in the cytoplasm.
  • Post-translational modifications include acetylation, phosphorylation, isopeptide bond (Ubl conjugation), and other modifications that may regulate its activity.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant fusion protein covering the full-length human DDX5 sequence (amino acids 1-614). This design may provide broad epitope recognition, but cross-reactivity with other DEAD box family members should be considered and validated in the relevant sample system.
  • For Western blotting, the expected molecular weight of DDX5 is approximately 69 kDa; a band at this size should be observed in positive samples. Endogenous levels may vary among cell lines and tissues.
  • In immunohistochemistry (paraffin-embedded sections), antigen retrieval conditions and antibody dilution should be optimized for specific tissue types, as localization is predominantly nuclear but may also be cytoplasmic.
  • For ELISA applications, the antibody can be used as a capture or detection reagent; pairing with another anti-DDX5 antibody may be necessary for sandwich ELISA development.
  • Cross-reactivity with mouse and rat is reported, making it suitable for studies in rodent models. However, sequence alignment and preliminary testing are recommended to confirm reactivity.
