Biological Background: DDX4 Function and Localization

  • DDX4, also known as VASA, is a member of the DEAD-box RNA helicase family and functions as a probable ATP-dependent RNA helicase.
  • It is essential for spermatogenesis, playing a critical role in germ cell development and differentiation (PubMed:10920202, PubMed:21034600).
    Related references: PMID:10920202, PMID:21034600
  • DDX4 is involved in transposon silencing via the piRNA pathway, helping to maintain genomic integrity in germ cells (By similarity).
  • It participates in secondary piRNA biogenesis, including PIWIL2-triggered piRNA production, utilizing its helicase activity for processing pre-piRNAs (By similarity).
  • The protein localizes to the cytoplasm and perinuclear region.
  • Expression is highly tissue-specific, found only in ovary and testis, and in migratory primordial germ cells.
  • DDX4 is regulated by phosphorylation and subject to alternative splicing.
  • Belongs to a broad range of biological processes including developmental protein, differentiation, meiosis, and RNA-mediated gene silencing.

Experimental Guidance and Technical Tips

  • The antibody was generated against a recombinant fragment covering amino acids 635–724 at the C-terminus of human DDX4. This region is likely conserved and may detect multiple isoforms.
  • For Western blot, a band at approximately 79 kDa is expected. Validate with appropriate positive controls from testis or ovary lysates, using relevant species (human, mouse, rat).
  • For immunofluorescence (IF-P) and immunohistochemistry (IHC-P), expect cytoplasmic and perinuclear staining. Use appropriate fixation and permeabilization, and consider antigen retrieval for IHC-P.
  • Cross-reactivity with mouse and rat was demonstrated, enabling use in these model organisms. Optimize antibody dilutions and conditions for each application and species.
  • Due to the restricted expression in germ cells, ensure tissue specificity during assay development.
  • This polyclonal antibody may be suitable for ELISA-based detection, but performance should be validated in the desired sandwich or indirect format.
