Experimental Guidance and Technical Tips

• For Western blotting, a band of ~73 kDa is expected; validate specificity using DDX3 knockout or knockdown lysates.
• In immunofluorescence, DDX3 shows dynamic localization (nucleus, cytoplasm, stress granules, etc.). Use appropriate fixation and permeabilization conditions; consider positive controls such as stressed cells to induce stress granule formation.
• For immunoprecipitation, the antibody is validated; ensure lysis buffer compatibility and include proper controls.
• For ELISA, the antibody can be used as a capture or detection reagent; determine optimal coating concentrations experimentally.
