Biological Background: ATP-dependent DNA helicase DDX11 Function and Localization
- DDX11 is an ATP-dependent DNA helicase that unwinds DNA in the 5' to 3' direction, requiring a minimal 5' single-stranded tail (15 nt for flaps, 10 nt for gapped substrates) for loading and translocation. It can displace duplex regions up to 100 bp, extendable to 500 bp by RPA or Ctf18-RFC. Related references: PMID:10648783, PMID:21854770, PMID:23797032, PMID:18499658, PMID:22102414
- It participates in DNA double-strand break repair at replication forks, stimulates FEN1 flap endonuclease activity to process Okazaki fragments, and associates with chromatin at replicating regions. Related references: PMID:23797032, PMID:18499658, PMID:27477908
- DDX11 cooperates with TIMELESS to preserve replication fork progression under stress and to regulate sister chromatid cohesion and mitotic chromosome segregation. Related references: PMID:26503245, PMID:17105772, PMID:18499658, PMID:20124417
- Its function at replication forks is modulated by the lncRNA DDX11-AS1/CONCR, which enhances ATPase activity and DNA binding. Related references: PMID:27477908
- DDX11 contributes to heterochromatin organization and activates rRNA transcription by recruiting UBTF and RNA polymerase I to hypomethylated rDNA loci. Related references: PMID:21854770, PMID:26089203
- The protein localizes to the nucleus, nucleolus, spindle poles, and midbody. It is highly expressed in spleen, thymus, testis, and melanoma cells, but not in normal melanocytes, and supports melanoma cell proliferation. Related references: PMID:23116066, PMID:9013641
- DDX11 is a phosphoprotein that coordinates iron-sulfur (4Fe-4S) clusters and contains ATP-binding and nucleic acid-binding motifs.
Experimental Guidance and Technical Tips
- For Western blot detection of endogenous DDX11 (~108 kDa), consider using whole-cell lysates from high-expression cell lines (e.g., melanoma lines) and ensure adequate gel separation for large proteins.
- Validate antibody specificity by including appropriate positive and negative controls, as polyclonal antibodies may exhibit batch-dependent variations.
- In ELISA, the recombinant immunogen (N-terminal 1–220 amino acids) can serve as a positive control or coating antigen; titrate antibody concentrations for optimal signal-to-noise ratios.
- Given the antibody's reactivity with human DDX11, cross-reactivity with orthologs or isoforms should be experimentally confirmed if used in other species.
