Biological Background: DDX1 Function and Localization
- DDX1 is an ATP-dependent RNA helicase, also known as DEAD box protein 1 or DEAD box protein retinoblastoma (DBP-RB).
- Acts as an ATP-dependent RNA helicase that unwinds RNA-RNA and RNA-DNA duplexes; possesses 5' single-stranded RNA overhang nuclease activity and ATPase activity on various RNA polynucleotides.
- May play a role in RNA clearance at DNA double-strand breaks (DSBs), facilitating template-guided repair of transcriptionally active genomic regions.
- Together with RELA, acts as a coactivator to enhance NF-kappa-B-mediated transcriptional activation; positively regulates CCND2 expression by binding to its promoter.
- May be involved in 3'-end cleavage and polyadenylation of pre-mRNAs; is an accessory subunit of the tRNA-splicing ligase complex that joins spliced tRNA halves. This step requires cooperation with ZBTB8OS for guanylylation of RTCB. Related references: PMID:24870230
- Component of a multi-helicase-TICAM1 complex that acts as a cytoplasmic sensor of viral dsRNA, activating antiviral responses including pro-inflammatory cytokine induction. Binds poly(I:C) dsRNA.
- Found in the nucleus, cytoplasm, cytoplasmic granules, cytosol, and mitochondria.
- Highest expression in retinoblastoma cell lines and neuroectodermal tissues (retina, brain, spinal cord).
