Biological Background: DDR1/MCK10/CD167a Function and Localization
- Epithelial discoidin domain-containing receptor 1 (DDR1) is a receptor tyrosine kinase that belongs to the discoidin domain receptor family. It is also known as CD167 antigen-like family member A, cell adhesion kinase, mammary carcinoma kinase 10 (MCK-10), and several other aliases. The protein undergoes alternative splicing to generate multiple isoforms.
- DDR1 functions as a cell surface receptor for fibrillar collagen and regulates cell attachment to the extracellular matrix, extracellular matrix remodeling, cell migration, differentiation, survival, and proliferation. Collagen binding triggers a signaling pathway involving SRC and leading to MAP kinase activation.
- The receptor promotes remodeling of the extracellular matrix by up-regulating matrix metalloproteinases MMP2, MMP7, and MMP9, thereby facilitating cell migration and wound healing. It also contributes to smooth muscle cell migration and arterial wound healing, and plays a role in tumor cell invasion.
- DDR1 is required for normal blastocyst implantation during pregnancy, normal mammary gland differentiation, and lactation. It is also necessary for normal ear morphology and hearing.
- Subcellularly, DDR1 is a single-pass type I membrane protein localized to the cell membrane, but a secreted form can also be generated.
- Key post-translational modifications include phosphorylation (the protein is autophosphorylated on tyrosine residues), N-linked glycosylation, and disulfide bond formation. It binds calcium and ATP.
- DDR1 is expressed at low levels in most adult tissues, with highest expression in brain, lung, placenta, and kidney. It is detected in various carcinoma cell lines, including breast, epidermoid, colon, and melanoma cells. In tissues, it is found in epithelia (e.g., colonic mucosa, thyroid follicles, pancreatic islets, kidney distal tubules) but not in connective tissue, endothelial cells, adipose tissue, muscle cells, or hematopoietic cells.
Experimental Guidance and Technical Tips
- The immunogen used comprises amino acids 21-416 of human DDR1, corresponding to the extracellular discoidin domain and part of the juxtamembrane region. Consider this targeting when designing assays that require specific domain recognition.
- For Western blot (WB), use reducing conditions and expect a band around 101 kDa. Validate the band identity in your specific sample type, as glycosylation and alternative splicing may produce additional bands.
- For ELISA, this monoclonal antibody can be used as a capture or detection reagent; however, optimal concentrations must be determined experimentally by the end user.
- Cross-reactivity with mouse and rat DDR1 has been demonstrated; therefore, this antibody is suitable for studies in rodent model systems. Always verify performance in the intended species and tissue context.
- As a monoclonal antibody, it provides batch-to-batch consistency, making it suitable for inclusion in standardized immunoassays.
