Biological Background: DDOST Function and Localization
- Protein identity and isoforms: DDOST encodes the dolichyl-diphosphooligosaccharide--protein glycosyltransferase 48 kDa subunit, also designated OST48, WBP1, AGER1, and CDG1R. Alternative splicing produces multiple isoforms.
- Core glycosylation function: The protein is an essential subunit of the oligosaccharyl transferase (OST) complex, which catalyzes the en bloc transfer of a preassembled high-mannose glycan (Glc3Man9GlcNAc2) from dolichol-pyrophosphate to asparagine residues in nascent polypeptides within the ER (PubMed:31831667).
Related references:
PMID:31831667 - Cotranslational mechanism: OST activity is coupled to protein translocation through the Sec61 translocon, enabling N-glycosylation to occur cotranslationally on the luminal side of the ER membrane.
- OST complex assembly: DDOST is required for the assembly of both STT3A- and STT3B-containing OST complexes, which mediate co‑ and post‑translational glycosylation, respectively (PubMed:22467853).
Related references:
PMID:22467853 - Subcellular localization: The protein is an integral membrane component of the endoplasmic reticulum with a signal peptide and multiple transmembrane domains.
- Clinical relevance: Mutations in DDOST are associated with congenital disorders of glycosylation, highlighting its importance in human health and disease.
- Molecular weight: The canonical DDOST polypeptide has a calculated molecular mass of approximately 50.8 kDa (UniProt), while the mature protein migrates at around 51 kDa on SDS‑PAGE.
Experimental Guidance and Technical Tips
- The immunogen (recombinant protein covering amino acids 40–300) corresponds to the luminal domain of human DDOST, likely generating antibodies that recognize both native and denatured epitopes. For WB, denaturing and reducing conditions are appropriate.
- When performing IHC-P, antigen retrieval with heat or enzymatic treatment may enhance detection; inclusion of a known positive control tissue such as liver or pancreas is recommended.
- For ELISA, the antibody may be used for direct or capture detection of recombinant or endogenous DDOST in cell and tissue lysates. Cross-reactivity with mouse and rat DDOST has been reported, so titration experiments in each species are advised.
- Signal optimization may require empirical determination of antibody dilution. Start with 1:500–1:2000 for WB and 1:200 for IHC-P and adjust as needed.
- Since DDOST is a ubiquitously expressed ER-resident protein, it can serve as a loading control or reference standard for glycosylation studies.
