Biological Background: DDIT3/CHOP Function and Localization

  • DDIT3 (DNA damage-inducible transcript 3), also designated CHOP (C/EBP-homologous protein), CHOP-10, and GADD153, is a member of the C/EBP family of transcription factors that acts as a dominant-negative inhibitor of C/EBP-mediated transcription.
  • It functions as a central mediator of the endoplasmic reticulum (ER) stress response, triggering cell cycle arrest and apoptosis under diverse stress conditions. Related references: PMID:15322075 PMID:15775988 PMID:19672300
  • DDIT3 heterodimerizes with other C/EBP proteins, impairing their DNA-binding capacity and repressing target gene expression.
  • As a transcriptional regulator, it induces pro-apoptotic genes such as TRIB3, BBC3/PUMA, and BCL2L11/BIM, while suppressing anti-apoptotic BCL2 and MYOD1 expression. Related references: PMID:15775988 PMID:17709599 PMID:20876114 PMID:18940792 PMID:19672300 PMID:20829347
  • DDIT3 negatively regulates canonical Wnt signaling by binding to TCF7L2/TCF4 and blocking its transcriptional activity. Related references: PMID:16434966
  • The protein dynamically shuttles between the cytoplasm and nucleus, accumulating in the nucleus upon ER stress to orchestrate gene expression programs.
  • DDIT3 is subject to post-translational modifications including phosphorylation and ubiquitin-like conjugation, which may modulate its activity and turnover.
  • Beyond apoptosis, DDIT3 participates in inflammatory responses by promoting caspase-11 (CASP4)-dependent IL-1β maturation and contributes to postnatal vascularization via NOS3 regulation.

Experimental Guidance and Technical Tips

  • The immunogen used to generate this antibody corresponds to the N-terminal region (amino acids 1–100) of human DDIT3, a domain that is relatively conserved; cross-reactivity with mouse and rat has been confirmed experimentally, but sequence alignment should be checked for other species.
  • For western blot (WB): detect a band at approximately 19 kDa; note that post-translational modifications or stress-induced isoforms may produce additional bands. Optimize blocking and antibody dilution to minimize background, as polyclonal preparations may exhibit non-specific binding.
  • For immunohistochemistry (IHC-P): antigen retrieval is recommended; validate staining in ER-stressed or apoptotic tissue sections where DDIT3 is upregulated.
  • For immunoprecipitation (IP): the antibody binds a linear peptide epitope and should effectively capture native DDIT3; include proper positive and negative controls to confirm specificity.
  • For ELISA: suitable for quantifying DDIT3 in cell lysates or serum samples; pair with a matched detection antibody if developing a sandwich assay.
  • As a polyclonal antibody, lot-to-lot consistency may vary; consider reserving a reference lot for long-term studies.
