Biological Background: DDIT3/CHOP Function and Localization
- DDIT3 (DNA damage-inducible transcript 3), also designated CHOP (C/EBP-homologous protein), CHOP-10, and GADD153, is a member of the C/EBP family of transcription factors that acts as a dominant-negative inhibitor of C/EBP-mediated transcription.

- DDIT3 heterodimerizes with other C/EBP proteins, impairing their DNA-binding capacity and repressing target gene expression.

- For western blot (WB): detect a band at approximately 19 kDa; note that post-translational modifications or stress-induced isoforms may produce additional bands. Optimize blocking and antibody dilution to minimize background, as polyclonal preparations may exhibit non-specific binding.
- For immunohistochemistry (IHC-P): antigen retrieval is recommended; validate staining in ER-stressed or apoptotic tissue sections where DDIT3 is upregulated.
