Biological Background: DDIT3/CHOP Function and Localization
- DDIT3, also known as CHOP, is a DNA damage-inducible transcript 3 protein and a member of the C/EBP family of transcription factors, functioning as a dominant-negative regulator and a stress-induced activator.
- It plays a central role in the ER stress response, inducing cell cycle arrest and apoptosis upon unresolved ER stress. Related references: PMID:15322075, PMID:15775988, PMID:19672300
- As a transcription factor, DDIT3 positively regulates pro-apoptotic and stress-related genes including TRIB3, BBC3/PUMA, and BCL2L11/BIM, while negatively regulating anti-apoptotic BCL2 and cell differentiation factors like MYOD1. Related references: PMID:15775988, PMID:17709599, PMID:20876114, PMID:18940792, PMID:19672300, PMID:20829347
- It inhibits the canonical Wnt signaling pathway by binding to TCF7L2/TCF4, impairing its DNA-binding and transcriptional activity. Related references: PMID:16434966
- DDIT3 localizes to both the cytoplasm and nucleus, consistent with its roles in signal transduction and gene regulation.
- Post-translational modifications include phosphorylation and ubiquitin-like conjugation, which may modulate its activity and stability.
- It is implicated in chromosomal rearrangements associated with myxoid liposarcoma, where a fusion with FUS results in an oncogenic transcription factor.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-100) of human DDIT3; consider using the immunizing peptide as a blocking control in relevant assays.
- For Western blotting, a band of approximately 19 kDa is expected; verify expression in ER stress-induced samples.
- For IHC-P and IF/ICC, optimize antigen retrieval and fixation conditions according to the sample type.
- Cross-reactivity with mouse and rat is reported; validate in the appropriate species model systems.
