Biological Background: DDIT3/CHOP Function and Localization
- DNA damage-inducible transcript 3 (DDIT3), also known as CHOP, C/EBP zeta, and GADD153, is a multifunctional transcription factor that belongs to the C/EBP family.
- Plays an essential role in the endoplasmic reticulum (ER) stress response, inducing cell cycle arrest and apoptosis under stress conditions. Related references: PMID:15322075, PMID:15775988
- Acts as a dominant-negative regulator of C/EBP-induced transcription by forming dimers with C/EBP family members, preventing their DNA binding and inhibiting target gene expression.
- Positively regulates the transcription of pro-apoptotic and inflammatory genes, including TRIB3, TNFRSF10B/DR5, BBC3/PUMA, BCL2L11/BIM, IL6, IL8, and IL23. Related references: PMID:15775988, PMID:17709599, PMID:20876114
