Biological Background: DDIT3/CHOP Function and Localization

  • DNA damage-inducible transcript 3 (DDIT3), also known as CHOP, C/EBP zeta, and GADD153, is a multifunctional transcription factor that belongs to the C/EBP family.
  • Plays an essential role in the endoplasmic reticulum (ER) stress response, inducing cell cycle arrest and apoptosis under stress conditions. Related references: PMID:15322075, PMID:15775988
  • Acts as a dominant-negative regulator of C/EBP-induced transcription by forming dimers with C/EBP family members, preventing their DNA binding and inhibiting target gene expression.
  • Positively regulates the transcription of pro-apoptotic and inflammatory genes, including TRIB3, TNFRSF10B/DR5, BBC3/PUMA, BCL2L11/BIM, IL6, IL8, and IL23. Related references: PMID:15775988, PMID:17709599, PMID:20876114
  • Negatively regulates the expression of anti-apoptotic BCL2 and differentiation factor MYOD1, and inhibits ATF4- and CEBPA-dependent transcriptional activation. Related references: PMID:18940792, PMID:19672300, PMID:20829347
  • Localizes to both the cytoplasm and nucleus, consistent with its dual role in cytoplasmic stress signaling and nuclear gene regulation.
  • Post-translationally modified by phosphorylation and ubiquitin conjugation, which may regulate its stability and activity.
  • Additionally inhibits the canonical Wnt signaling pathway by binding to TCF7L2/TCF4 and repressing its transcriptional activity. Related references: PMID:16434966

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-100) of human DDIT3; therefore, the antibody likely recognizes an epitope within this domain.
  • For Western blotting, a band near 19 kDa is expected, but consider that post-translational modifications may cause slight shifts in apparent molecular weight.
  • In immunohistochemistry (paraffin-embedded tissues), optimize antigen retrieval and validate staining in tissues with known ER stress or apoptosis induction, such as treated cancer cell lines or disease models.
  • For ELISA, ensure the coating peptide or protein is within the N-terminal 1-100 region to capture the antibody effectively; cross-reactivity with rodent orthologs may be leveraged for multi-species assays.
  • As with any antibody, performance should be validated in the end-user's specific sample system and experimental conditions.
