  • DDB2 (DNA damage-binding protein 2) is a core component of the UV-DDB complex, which recognizes UV-induced DNA lesions such as cyclobutane pyrimidine dimers and 6-4 photoproducts, thereby recruiting nucleotide excision repair (NER) machinery.   • It also serves as the substrate recognition module for the DDB2-CUL4-ROC1 E3 ubiquitin ligase complex, mediating ubiquitination of histones H2A, H3, and H4 at damage sites to facilitate chromatin remodeling and repair. Related references: PMID:12732143, PMID:16473935, PMID:16678110
  • This complex additionally ubiquitinates XPC, enhancing its DNA-binding and promoting NER, and targets KAT7/HBO1 for degradation upon DNA damage. Related references: PMID:15882621, PMID:26572825
