Biological Background: DDB1 Function and Localization

  • DDB1 (DNA damage-binding protein 1), also known as XPE, DDBA, XAP1, and UV-DDB1, is a 127 kDa protein that serves as a core component of the UV-DDB complex, which recognizes UV-induced DNA lesions such as cyclobutane pyrimidine dimers (CPD) and 6-4 photoproducts to initiate nucleotide excision repair (NER).
    Related references: PMID:15448697, PMID:16260596, PMID:16407242

  • DDB1 is a substrate adaptor for multiple DCX (DDB1-CUL4-X-box) E3 ubiquitin-protein ligase complexes, which mediate ubiquitination and subsequent proteasomal degradation of target proteins, with functional specificity dictated by the variable substrate recognition component recruited.
    Related references: PMID:14739464, PMID:16407252, PMID:16482215

  • Through DCX(DDB2) complex, DDB1 facilitates the ubiquitination of histones H2A, H3, and H4 at UV-damaged sites, promoting histone removal from nucleosomes to facilitate DNA repair.
    Related references: PMID:16473935, PMID:16678110, PMID:17041588

  • DDB1 is also involved in the ubiquitination of XPC, enhancing its DNA-binding activity and promoting NER, and participates in transcription-coupled repair (TCR) via the DCX(ERCC8) complex.
    Related references: PMID:15882621, PMID:12732143, PMID:32355176, PMID:38316879

  • The DDB1-CUL4A-DTL E3 ligase complex regulates circadian clock function by mediating the ubiquitination and degradation of CRY1, and influences gluconeogenesis through FOXO1 stabilization.
    Related references: PMID:26431207

  • Subcellular localization includes both cytoplasm and nucleus, reflecting its roles in DNA repair signaling and protein degradation pathways.

  • DDB1 is subject to post-translational modifications including acetylation, phosphorylation, and ubiquitination, and is implicated in viral interactions and intellectual disability-associated variants.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 1000-1140 in the C-terminal region of human DDB1. For western blotting (WB), a band at approximately 127 kDa is expected; consider the possibility of detecting splice isoforms if the epitope is conserved.
  • For immunohistochemistry on paraffin sections (IHC-P), antigen retrieval conditions may need optimization due to the large protein size and potential nuclear localization.
  • When performing immunoprecipitation (IP), antibody concentration and incubation time should be empirically determined, as polyclonal preparations may vary between lots.
  • In ELISA, the antibody can be used as a capture or detection reagent; validate cross-reactivity against mouse and rat DDB1 if working with multispecies samples.
