Biological Background: DDAH2 Function and Localization

  • DDAH2 (Protein G6a, DDAHII) is an inactive member of the dimethylarginine dimethylaminohydrolase family that does not metabolize N(G),N(G)-dimethyl-L-arginine (ADMA) (PubMed:21493890, 37296100). Related references: PMID:21493890 PMID:37296100
  • In endothelial cells, it induces vascular endothelial growth factor (VEGF) expression through PKA-dependent phosphorylation of transcription factor SP1, independent of nitric oxide (NO) and NO synthase (By similarity).
  • It enhances pancreatic insulin secretion by upregulating secretagogin (SCGN), an insulin vesicle docking protein, via SP1-mediated transcriptional activation (By similarity).
  • Upon viral infection, DDAH2 translocates to mitochondria and triggers mitochondrial fission through activation of DNM1L, thereby inhibiting MAVS-dependent innate immune responses (PubMed:33850055). Related references: PMID:33850055
  • The protein localizes to both the cytoplasm and mitochondria.
  • DDAH2 is widely expressed, with detection in heart, placenta, lung, liver, skeletal muscle, kidney, and pancreas, and at very low levels in brain.
