Biological Background: DCTN2 Function and Localization
- DCTN2 (also known as dynamitin, p50, RBP50, DCTN50) is a subunit of the dynactin complex, a key activator of the molecular motor dynein.
- It functions in ultra-processive transport along microtubules, binding to the ACTR1A filament within the dynactin shoulder domain and acting as a molecular ruler to determine dynactin length.
- DCTN2 modulates cytoplasmic dynein binding to organelles and plays a critical role in prometaphase chromosome alignment and spindle organization during mitosis.
- It is involved in anchoring microtubules to centrosomes, contributing to cytoskeletal organization and cell division.
- The protein is localized to the cytoplasm, cytoskeleton, microtubule organizing center (centrosome), and membrane.
- DCTN2 is subject to post-translational modifications including acetylation and phosphorylation, and the gene undergoes alternative splicing.
- May also play a role in synapse formation during brain development.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody spans a region within the coiled-coil domain of DCTN2, which may provide specificity for the native protein.
- For western blot (WB), validate performance in the appropriate cell or tissue lysate; a band near 44 kDa is expected.
- For ELISA, consider using recombinant DCTN2 as a positive control to confirm reactivity.
- This antibody cross-reacts with human, mouse, and rat DCTN2; however, optimal dilution and conditions should be determined in the relevant sample system.
