Biological Background: DCP2 Function and Localization
- DCP2 (NUDT20) is an m7GpppN-mRNA hydrolase, also known as nucleoside diphosphate-linked moiety X motif 20, that functions as a decapping metalloenzyme and is also referred to as hDpc or Nudix motif 20.
- DCP2 catalyzes the cleavage of the 7-methyl guanine cap structure on mRNA, yielding a 5'-phosphorylated mRNA fragment and 7m-GDP (PubMed:12486012, PubMed:12923261, PubMed:21070968). It has higher activity towards non-polyadenylated mRNAs and requires an RNA moiety for cap recognition (PubMed:21070968).
Related references: PMID:12486012, PMID:12923261, PMID:21070968 - DCP2 is necessary for both normal mRNA turnover and nonsense-mediated mRNA decay (PubMed:14527413).
Related references: PMID:14527413 - It plays a role in replication-dependent histone mRNA degradation (PubMed:18172165).
Related references: PMID:18172165 - The presence of a N6-methyladenosine (m6A(m)) cap at the second transcribed position provides resistance to DCP2-mediated decapping (PubMed:28002401).
Related references: PMID:28002401 - DCP2 blocks autophagy under nutrient-rich conditions by repressing ATG-related gene expression through transcript degradation (PubMed:26098573).
Related references: PMID:26098573 - Subcellularly, DCP2 is found in the cytoplasm, P-bodies, and nucleus. It is expressed in brain and testis but not detected in heart at the protein level.
- Keywords associated with DCP2 include alternative splicing, metal-binding, phosphoprotein, and RNA-binding, reflecting its regulation and interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-240) of human DCP2. This polyclonal antibody may recognize multiple isoforms due to alternative splicing; validation in the relevant sample system is recommended.
- For Western blot, a band around 48 kDa (predicted molecular weight) is expected. Given DCP2's expression in brain and testis, lysates from these tissues or cell lines derived from them are suitable positive controls.
- In ELISA, the antibody may be used for detecting recombinant or endogenous DCP2; consider optimizing coating and detection conditions.
- As a polyclonal antibody, cross-reactivity with other Nudix family members is possible; inclusion of appropriate controls is advised.
