Biological Background: DCLK1 Function and Localization

  • DCLK1 (Doublecortin-like kinase 1) is a serine/threonine-protein kinase containing doublecortin domains, implicated in calcium-signaling pathways controlling neuronal migration in the developing brain.
  • It may also participate in functions of the mature nervous system.
  • In fetal tissues, DCLK1 is highly expressed in the brain, and detectable in lung and liver; it is not expressed in fetal kidney.
  • In adult tissues, it is ubiquitously expressed in the brain and detectable in heart, liver, spleen, thymus, prostate, testis, ovary, small intestine, and colon.
  • Alternative splicing generates type A isoforms, predominantly expressed in fetal brain, and type B isoforms, present in both fetal and adult brain.
  • Classified as a developmental protein, it is involved in neurogenesis and differentiation.
  • The protein is a phosphoprotein and binds ATP, consistent with its kinase activity.

Experimental Guidance and Technical Tips

  • For Western Blot (WB), expect a band around 82 kDa. Use lysates from neuronal tissues or DCLK1-expressing cell lines, and validate loading controls.
  • For Immunofluorescence-Paraffin (IF-P), optimize fixation and antigen retrieval conditions for your sample type, particularly in brain or embryo tissues.
  • Since this is a polyclonal antibody, perform lot-specific dilution optimization for each application (e.g., ELISA) to ensure consistent signal-to-noise ratios.
  • Cross-reactivity with mouse and rat indicates suitability for models; however, verify band size and specificity in your experimental system.
  • The immunogen corresponds to amino acids 590-740 near the C-terminus; epitope accessibility may vary depending on protein conformation and sample preparation.
