Biological Background: DCAF12 Function and Localization
- DCAF12 (DDB1- and CUL4-associated factor 12), also known as CT102, TCC52, WDR40A, is a WD repeat-containing protein.
- It functions as a substrate-recognition component of the DCX (DDB1-CUL4-X-box) E3 ubiquitin ligase complex, specifically recognizing C-terminal degrons (C-degrons) to target proteins for ubiquitination and degradation. Related references: PMID:16949367, PMID:16964240, PMID:29779948
- The DCX(DCAF12) complex recognizes diglutamate (Glu-Glu) C-termini in substrates such as MAGEA3, MAGEA6, and CCT5, leading to their degradation; this activity is required for starvation-induced autophagy. Related references: PMID:29779948, PMID:31267705
- DCAF12 also recognizes an alternative glutamate-leucine (Glu-Leu) C-degron, as seen in MOV10, and controls MOV10 protein levels in spermatogenesis and activated T cells. Related references: PMID:34065512
