Biological Background: DC-SIGNR/CD299 Function and Localization

  • DC-SIGNR/CD299 (C-type lectin domain family 4 member M) is also known as L-SIGN, DC-SIGN2, and CD209L. It belongs to the C-type lectin receptor family and shares homology with DC-SIGN (CD209).
  • Functions as a pathogen-recognition receptor involved in peripheral immune surveillance, particularly in the liver. It mediates endocytosis of pathogens for lysosomal degradation.
  • Acts as an attachment receptor for numerous viruses, including HIV-1, hepatitis C virus, SARS-CoV, Ebola virus, influenza virus, and others, facilitating viral entry into host cells.
  • Recognizes ICAM3 via mannose-like carbohydrate binding, contributing to dendritic cell-T cell interactions.
  • Localizes to the cell membrane and can also be secreted. The secreted form is produced by alternative splicing.
  • Predominantly expressed in liver sinusoidal endothelial cells and lymph node, with additional expression in placental endothelium, type II alveolar cells, and lung endothelial cells. Not found in macrophages.
  • Post-translational modifications include N-linked glycosylation and disulfide bond formation, which are important for ligand binding and structural stability. Its molecular weight varies due to glycosylation, observed between 24 and 45 kDa.

  • The immunogen spans the extracellular domain (aa 71-399), enabling detection of both membrane-bound and secreted DC-SIGNR isoforms. For Western blot, validate in liver endothelial cell lines as positive controls; the glycosylated protein migrates between 24-45 kDa.
  • For IF/ICC, expect pronounced cell surface staining; permeabilization may reveal intracellular pools. For flow cytometry, use live cells for surface expression or fixed/permeabilized cells for total detection.
  • In ELISA applications, the antibody can serve as capture or detection reagent; include recombinant protein standards for quantitation.
  • Cross-reactivity is restricted to human; confirm suitability in your assay system when working with other species.
