- DBNL (Drebrin-like protein), also known as HIP-55, SH3P7, or ABP1, is an adapter protein encoded by the DBNL gene. It exists in multiple isoforms due to alternative splicing.
- It binds F-actin and dynamin-1 (DNM1) and plays a role in receptor-mediated endocytosis and actin cytoskeleton reorganization. It does not bind G-actin and does not promote actin polymerization on its own.
- DBNL is involved in the formation of cell projections, such as neurites, and contributes to neuron morphogenesis and synapse formation via interactions with WASL and COBL.
- Required for the assembly of organized podosome rosettes, actin-rich structures important for adhesion and invasion.
- In leukocytes, it acts as a common effector of antigen receptor-signaling pathways and is a key component of the immunological synapse, bridging T-cell receptor (TCR) signaling to the actin cytoskeleton and gene activation.
- Subcellularly, DBNL localizes to the cytoplasm, cytoskeleton, cell cortex, lamellipodia, ruffles, podosomes, clathrin-coated vesicles, early endosomes, Golgi apparatus, and postsynaptic densities.
- The protein is regulated by acetylation and phosphorylation, and it is expressed in tissues with prominent roles in the nervous and immune systems.
