Biological Background: DBI Function and Localization

  • DBI encodes the Acyl-CoA-binding protein (ACBP), also known as diazepam-binding inhibitor (DBI) and endozepine (EP).
  • It is a lipid-binding protein that binds medium- and long-chain acyl-CoA esters with very high affinity, functioning as an intracellular acyl-CoA carrier.
  • DBI can displace diazepam from the benzodiazepine recognition site on the GABA type A receptor, suggesting a role as a neuropeptide in modulating GABAergic neurotransmission.
  • Subcellularly, DBI localizes to the endoplasmic reticulum and Golgi apparatus, consistent with its role in lipid transport and metabolism.
  • Multiple isoforms are produced via alternative promoters and splicing, with isoform 1 ubiquitously expressed, isoform 2 highly enriched in liver and adipose tissue, and isoform 3 strongly expressed in heart and adipose tissue.
  • Post-translational modifications include acetylation, hydroxylation, and phosphorylation, which may regulate its activity and localization.
  • DBI belongs to the acyl-CoA-binding protein family and is involved in transport; it is a proteomics-identified reference proteome component.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1-104) of human and mouse DBI, which is highly conserved; this supports the observed cross-reactivity with rat.
  • For Western blot (WB), the predicted protein weight is 10-12 kDa; consider using high-percentage gels and optimized transfer conditions for small proteins.
  • For immunohistochemistry (IHC-P), antigen retrieval and detection conditions should be tailored to the tissue type, given the moderate expression of isoform 1 and tissue-specific enrichment of other isoforms.
  • Validate the antibody in your relevant sample system, as endogenous expression levels may vary across tissues and species.
