Biological Background: DBI Function and Localization
- DBI (Diazepam-binding inhibitor), also known as Acyl-CoA-binding protein (ACBP), endozepine (EP), or ACBD1, is a ~10 kDa protein encoded by the Dbi gene in mice.
- It binds medium- and long-chain acyl-CoA esters with very high affinity and is proposed to function as an intracellular carrier of acyl-CoA esters.
- DBI also acts as a neuropeptide: it can displace diazepam from the benzodiazepine recognition site on the GABA type A receptor, potentially modulating GABAergic neurotransmission.
- Subcellular localization is mainly to the endoplasmic reticulum and Golgi apparatus, consistent with its role in lipid metabolism and trafficking.
- The protein undergoes post-translational modifications including acetylation, hydroxylation, and phosphorylation, which may regulate its activity or localization.
- As a lipid-binding protein, DBI is involved in intracellular acyl-CoA transport, impacting processes such as fatty acid oxidation, membrane synthesis, and signal transduction.

- The immunogen used is a recombinant fusion protein corresponding to the N-terminal 1-87 amino acids of mouse DBI (NP_031856.1). This region is well-conserved; the antibody is expected to recognize the full-length endogenous protein.
- For Western blot, a band at 10-12 kDa is expected under reducing conditions. Validate with appropriate positive controls (mouse or rat tissue lysates known to express DBI).
- In immunofluorescence and immunohistochemistry (paraffin sections), DBI localizes to the ER and Golgi apparatus. Co‑staining with organelle markers is recommended to confirm specificity.
- For ELISA, the antibody can be used as a capture or detection reagent; pair with a monoclonal antibody for sandwich ELISA after validation in your assay system.
- Since DBI is ubiquitously expressed, most tissues should be positive, but consider testing a range of samples to establish baseline expression.
