Biological Background: DBI Function and Localization
- The DBI gene encodes Acyl-CoA-binding protein (ACBP), also known as Diazepam-binding inhibitor (DBI) and Endozepine (EP).
- ACBP binds medium- and long-chain acyl-CoA esters with high affinity, functioning as an intracellular carrier for these lipids.
- The protein may also act as a neuropeptide by displacing diazepam from the benzodiazepine recognition site on GABA type A receptors, potentially modulating GABAergic neurotransmission.
- DBI is localized to the endoplasmic reticulum and Golgi apparatus.
- Multiple isoforms exist: isoform 1 is expressed ubiquitously at moderate levels; isoform 2 is highly expressed in liver and adipose tissue; isoform 3 shows strong expression in adipose tissue and heart.
- Post-translational modifications include acetylation, hydroxylation, and phosphorylation, as indicated by proteomics data.
- The protein belongs to the acyl-CoA-binding protein family and is involved in lipid transport and metabolism.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human DBI protein (amino acids 1-104), suggesting the antibody recognizes the entire protein.
- For western blotting, a band of approximately 10-12 kDa is expected; consider using a high-percentage gel for better resolution of small proteins.
- In immunofluorescence/immunocytochemistry, staining consistent with endoplasmic reticulum and Golgi localization may be observed.
- The antibody cross-reacts with mouse and rat, allowing validation in rodent models; relevant tissues include liver, adipose tissue, and heart where DBI isoforms are abundant.
- When performing immunoprecipitation, confirm capture efficiency with a suitable detection antibody or by mass spectrometry.
- ELISA applications may require optimization of coating antigen and antibody dilution; include appropriate controls.
