Biological Background: DAZAP2 Function and Localization

  • DAZAP2 (DAZ-associated protein 2), also known as PRTB (proline-rich transcript in brain protein), is encoded by the DAZAP2 gene. Alternative splicing produces multiple isoforms.
  • In unstressed cells, it promotes SIAH1-mediated polyubiquitination and degradation of the serine/threonine-protein kinase HIPK2, likely by facilitating complex formation between HIPK2 and the E3 ligase SIAH1. Related references: PMID:33591310
  • Upon DNA damage, DAZAP2 is phosphorylated by HIPK2 and translocates to the nucleus, where it binds TP53/p53 at target gene promoters, modulating a subset of cell death-mediating genes to reduce apoptosis. Related references: PMID:33591310
  • DAZAP2 enhances the binding of transcription factor TCF7L2/TCF4, a Wnt pathway effector, to gene promoters, implicating it in Wnt signaling regulation.
  • It plays a role in stress granule formation, contributing to the cellular stress response. Related references: PMID:17984221
  • Subcellular localization includes the cytoplasm, nucleus, nuclear speckles, nuclear bodies, and stress granules, reflecting dynamic shuttling under different conditions.
  • DAZAP2 is widely expressed, with high levels in spleen, thymus, prostate, testis, ovary, small intestine, colon, and leukocytes; its expression is down-regulated in multiple myeloma.
  • Post-translational modifications include phosphorylation and ubiquitination, as highlighted by the keywords Phosphoprotein and Ubl conjugation.

Experimental Guidance and Technical Tips

  • The immunogen is the full-length human DAZAP2 sequence, which may detect both known isoforms and the orthologous mouse protein due to the reported cross-reactivity.
  • For Western blotting, the predicted molecular weight is approximately 17 kDa, but actual migration can be influenced by post-translational modifications; validate in the relevant sample system.
  • When performing ELISA, polyclonal antibodies may offer enhanced sensitivity through epitope multiplicity; optimal working titers should be determined empirically.
  • Since DAZAP2 participates in stress granule formation and DNA damage responses, consider appropriate cellular treatments to induce its nuclear translocation or expression changes.
