Biological Background: Daxx Function and Localization

  • Death domain-associated protein 6 (Daxx) is a ubiquitously expressed transcription corepressor that represses basal and activated transcription, particularly of sumoylated transcription factors. It modulates transcriptional activity by localizing to subnuclear compartments such as PML nuclear bodies and nucleolus.
  • Daxx acts as a histone chaperone for H3.3, facilitating replication-independent deposition of histone H3.3 at pericentric repeats and telomeres through the ATRX:DAXX chromatin remodeling complex. It recruits H3.3:H4 dimers to PML nuclear bodies, a process independent of ATRX but facilitated by ASF1A.
  • As an adapter protein, Daxx regulates the MDM2-DAXX-USP7 complex, balancing TP53 degradation under non-stress conditions and promoting TP53 stabilization upon DNA damage by disrupting MDM2 autoubiquitination.
  • Daxx is proposed to mediate JNK pathway activation and apoptosis via MAP3K5 in response to TNFRSF6 and TGFBR2 signaling, though this role may be context-dependent and inhibited by HSPB1/HSP27 interaction.

  • Subcellular localization includes cytoplasm, nucleoplasm, PML bodies, nucleolus, and centromeric chromatin. It is involved in the ectopic mislocalization of centromeric histone variant CENPA in tumors.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide spanning amino acids 200–300 of human Daxx; therefore, the antibody likely recognizes an epitope within this central region.
  • For Western blot (WB), consider using nuclear extracts or whole-cell lysates from human cell lines. Validate the observed band size against the predicted molecular weight of ~81 kDa.
  • In ELISA, direct coating with the immunizing peptide or recombinant Daxx protein may be used to assess antibody reactivity. Cross-reactivity with other Daxx isoforms or homologs should be tested experimentally.
