- DAB2 (Disabled homolog 2) is an adapter protein also known as adaptor molecule disabled-2, DOC-2, and differentially expressed in ovarian carcinoma 2.
- Functions as a clathrin-associated sorting protein (CLASP) essential for clathrin-mediated endocytosis; binds clathrin and PtdIns(4,5)P2 to recruit cargo containing NPXY motifs (e.g., LDL receptor) to coated pits.
- Involved in TGF-beta receptor signaling, facilitating SMAD2 phosphorylation; mediates TGF-beta-stimulated JNK activation and may inhibit canonical Wnt/beta-catenin signaling by stabilizing the destruction complex and sequestering LRP6.
- Localizes to the cytoplasm, associated with clathrin-coated pits and vesicles at the membrane.
- Expressed in ovarian surface epithelial cells, breast epithelial cells, spleen, thymus, prostate, testis, macrophages, fibroblasts, lung, placenta, brain stem, heart, small intestine, and kidney proximal tubular cells at the protein level.
- Subject to acetylation and phosphorylation; involved in apoptosis, differentiation, development, endocytosis, protein transport, and Wnt signaling; acts as a tumor suppressor.
- Required for visceral endoderm formation, cell positioning, and epithelial-to-mesenchymal transition during embryogenesis; may also promote myeloid cell differentiation and macrophage adhesion.
