Biological Background: DWARF 53 (D53) Function and Localization
- D53 functions as a repressor of strigolactone (SL) signaling, a phytohormone pathway critical for shoot branching and tillering in rice (PubMed:24336200, PubMed:24336215). Related references: PMID:24336200 PMID:24336215
- D53 suppresses the transcriptional activation activity of SPL14/IPA1, and together they mediate SL-regulated tiller development (PubMed:28809396). Related references: PMID:28809396
- D53 expression is negatively regulated by SPL14/IPA1, which binds to the D53 promoter to repress its transcription, forming a feedback loop in SL signaling (PubMed:28809396). Related references: PMID:28809396
- D53 localizes to the nucleus, consistent with its role as a transcriptional corepressor.
- According to UniProt annotations, D53 contains repeat sequences and is predicted to undergo ubiquitin-like conjugation, which may influence its stability or activity.
- D53 is expressed in shoot bases of seedlings, young leaves, axillary buds, and young panicles, as well as in the vasculature of young roots, culms, internodes, and nodes, preferentially in parenchyma cells surrounding the xylem (PubMed:24336200, PubMed:24336215). Related references: PMID:24336200 PMID:24336215
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 1000-1100 at the C-terminus of D53. This region may be used for epitope mapping or validation studies.
- For Western blotting, note that D53 has a predicted molecular weight of approximately 123 kDa. Use appropriate gel percentages and transfer conditions for high molecular weight proteins.
- In ELISA, titrate the antibody against the recombinant immunogen or native protein extracts from Oryza sativa to determine optimal working concentrations.
- Cross-reactivity has been demonstrated with Oryza sativa; validation in additional species or sample types should be performed if required.
