Biological Background: Cytokeratin AE1 (Type I Keratins) Function and Localization
- Keratin 10 (K10) encoded by KRT10 is a type I keratin and a major constituent of intermediate filaments in suprabasal keratinocytes of stratified epithelia.
- Forms heteropolymers primarily with type II keratin KRT1, essential for terminal differentiation, mechanical resilience, and epidermal barrier formation.
- Alternative names include Cytokeratin-10 (CK-10) and Keratin-10 (K10); associated with diseases such as ichthyosis.
- Subcellular localization observed in secreted, extracellular space, cell surface, and cytoplasm; expressed in all suprabasal cell layers including stratum corneum, and on the surface of lung cell lines and desquamated nasal epithelial cells.
- Keywords: Disulfide bond, phosphoprotein, coiled coil; undergoes post-translational modifications.
- Plays a role in microbial infection: mediates S. aureus adherence to nasal epithelial cells via clfB (PubMed:12427098) and binds S. pneumoniae PsrP, facilitating bacterial colonization of lung cells (PubMed:19627498). Related references: PMID:12427098, PMID:19627498
- The AE1 clone recognizes a broad spectrum of type I keratins including K10, K14, K15, K16, and K19, serving as a pan-epithelial marker.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to amino acids 101-200 of human K10. Validate the antibody’s reactivity in your target species, considering reported cross-reactivity with mouse and rat.
- For IHC-P and IF-P applications on paraffin-embedded tissues, optimize antigen retrieval conditions (e.g., heat-induced or protease-based retrieval) to ensure proper unmasking of the intermediate filament epitopes.
- For ELISA, consider using the antibody as a capture or detection reagent with appropriate blocking steps to minimize nonspecific interactions.
- Given the pan-keratin AE1 specificity, this antibody is well-suited for identifying epithelial cells and carcinomas. Include relevant controls to confirm subtype-level identification if needed.
- Perform initial dilution optimization for each application, as performance may vary depending on sample type and experimental conditions.
