Biological Background: Cytokeratin AE1 (KRT10) Function and Localization

  • Keratin 10 (KRT10) is a type I cytoskeletal keratin encoded by the KRT10 gene, also known as cytokeratin-10 (CK-10) or keratin-10 (K10). It belongs to the intermediate filament family and forms heterodimers with type II keratins.
  • KRT10 is a major structural component of intermediate filaments in suprabasal keratinocytes of stratified epithelia, where it partners primarily with KRT1 (and KRT2) to assemble filament networks that provide mechanical resilience, support terminal differentiation, and maintain the epidermal barrier.
  • Subcellularly, KRT10 localizes to the cytoplasm, but it can also be secreted and detected in the extracellular space and on the cell surface, particularly in differentiated keratinocytes and certain epithelial cells.
  • KRT10 is highly expressed in all suprabasal layers of the epidermis, including the stratum corneum, and is also found on the surface of lung cell lines and desquamated nasal epithelial cells. Related references: PMID:12427098 PMID:19627498
  • In microbial infections, KRT10 mediates adherence of Staphylococcus aureus to desquamated nasal epithelial cells via clfB, potentially facilitating nasal colonization, and also binds Streptococcus pneumoniae PsrP protein, promoting bacterial adherence to lung cells.
  • Post-translational modifications include phosphorylation, and structural features like coiled-coil domains and disulfide bonds contribute to filament stability. KRT10 mutations are associated with ichthyosis disorders.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 101-200 of human Cytokeratin AE1 (NP_000412.4). Given the polyclonal nature and pan-keratin reactivity, this antibody is suitable for detecting multiple keratins in human, mouse, and rat samples. For WB, consider the expected molecular weights: ~59 kDa (KRT10), 52 kDa (KRT14), 49 kDa (KRT15), 51 kDa (KRT16), and 44 kDa (KRT19). Validate in your system with appropriate controls.
  • For IHC-P, antigen retrieval may be required; test on known positive controls such as skin or other stratified epithelium.
  • In ELISA, ensure the coating peptide corresponds to the target keratins; cross-reactivity should be verified with individual recombinant keratin proteins.
  • Since this antibody reacts with multiple keratins, single-band specificity may not be achieved in WB without further characterization; consider using specific monoclonal antibodies for unambiguous identification.
