Biological Background: Cytokeratin 9 (KRT9) Function and Localization
- Cytokeratin 9 is a type I intermediate filament protein (keratin) primarily expressed in the suprabasal layers of the palmoplantar epidermis.
- It serves as a structural component, forming heteropolymers with type II keratins to assemble the keratin intermediate filament network, thereby maintaining the mechanical resilience of palm and sole skin.
- The protein is characterized by a coiled-coil domain and undergoes phosphorylation, which may regulate filament dynamics.
- Mutations in KRT9 are associated with epidermolytic palmoplantar keratoderma, an autosomal dominant skin disorder.
- Tissue expression is restricted to the terminally differentiated epidermis of palms and soles, marking it as a specific marker for these sites.
Experimental Guidance and Technical Tips
- For Western blot, a band around 62 kDa is expected; use denaturing and reducing conditions, and include appropriate positive controls such as lysates from palmoplantar skin or cell lines expressing KRT9.
- For ELISA, consider testing the antibody in both direct and sandwich formats; pair with a suitable type II keratin antibody for specific detection.
- Due to cross-reactivity with mouse and rat, verify reactivity in the relevant species samples; sequence alignment of the immunogen region (aa 254-412) shows high conservation among these species, but empirical validation is recommended.
- The antibody may detect phosphorylated and unphosphorylated forms; treat samples with phosphatase inhibitors if needed.
