Biological Background: Cytokeratin 7 Function and Localization
- Keratin 7 (KRT7) is a type II cytoskeletal keratin, a member of the intermediate filament family, encoded by the KRT7 gene (also known as SCL). Alternative names include CK-7, K7, sarcolectin, and type-II keratin Kb7.
- KRT7 blocks interferon-dependent interphase and stimulates DNA synthesis, thereby influencing cell proliferation.
- It is involved in the translational regulation of human papillomavirus type 16 (HPV16) E7 mRNA, implicating a role in host-virus interactions.
- Subcellular localization is primarily cytoplasmic, consistent with its function as an intermediate filament protein.
- Tissue expression is observed in cultured epidermal, bronchial, and mesothelial cells, but absent in colon, ectocervix, and liver; it is expressed in glandular cells of the gastroesophageal junction but not in the esophagus.
- The protein undergoes several post-translational modifications, including acetylation, phosphorylation, methylation, and ubiquitin conjugation (Ubl), as well as forming isopeptide bonds. It is also a glycoprotein.

- This rabbit monoclonal antibody was generated against a synthetic peptide corresponding to amino acids 1–100 of human KRT7 (P08729). The immunogen lies in the N-terminal region, which may be well-exposed in the native protein.
- For Western blotting (WB), denaturing and reducing conditions are recommended; a band at ~51 kDa is expected. Validate the antibody in a relevant cell or tissue lysate expressing KRT7.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), consider antigen retrieval methods (e.g., heat-induced epitope retrieval) and appropriate fixation to preserve epitope integrity. Validate in positive control tissues such as glandular epithelium or bronchial cells.
- The antibody shows cross-reactivity with mouse and rat KRT7 orthologs, as per supplier data, but cross-reactivity should be confirmed experimentally in the relevant species and application.
- As with any monoclonal antibody, lot-to-lot consistency should be monitored, and optimal working dilutions determined empirically for each assay.
