Biological Background: Cytokeratin 6 (KRT6) Function and Localization
- KRT6A (Cytokeratin 6A) is a type II keratin that forms obligatory heteropolymers with type I keratins KRT16 and KRT17, assembling into intermediate filament networks critical for maintaining the structural integrity of stratified epithelia, particularly in the epidermis, nail bed, and oral mucosa. - During wound healing, KRT6A is rapidly induced in keratinocytes at the wound edge and participates in cytoskeletal reorganization during re-epithelialization, facilitating the restoration of the epithelial barrier.
- KRT6A negatively regulates collective keratinocyte migration by stabilizing non-muscle myosin MYH9 and desmoplakin, altering cell adhesion dynamics, and by inhibiting SRC activity, which reduces the speed and directionality of epithelial sheet movement.

- The immunogen spans the N-terminal region (aa 1-100) of human KRT6A, which may confer isoform-specific recognition; however, due to high sequence homology among type II keratins, cross-reactivity with KRT6B, KRT6C, or other keratins should be evaluated in the relevant sample system.
- For Western blot, a band around 60 kDa is expected; consider using human epithelial cell lines (e.g., HaCaT, A431) or skin/corneal tissue lysates as positive controls.
- This polyclonal antibody is suitable for ELISA; for sandwich ELISA development, pairing with a monoclonal detection antibody is recommended, and optimal dilutions should be determined empirically.
