Biological Background: Cytokeratin 6 (KRT6) Function and Localization
- Keratin 6A (KRT6A) is a type II intermediate filament protein that forms obligate heteropolymers with type I keratins KRT16 and KRT17, essential for keratin cytoskeleton assembly.
- It provides structural integrity and stress resilience to stratified epithelia, including the epidermis, nail bed, and oral mucosa (PubMed:11886499, PubMed:17719747, PubMed:7545493).

PMID:7545493 - KRT6A is rapidly induced in wound-edge keratinocytes and facilitates cytoskeletal reorganization during re-epithelialization (By similarity).
- It negatively regulates collective keratinocyte migration by stabilizing non-muscle myosin MYH9 and desmoplakin, thereby modulating cell adhesion and movement (By similarity).
- KRT6A also limits epithelial migration by inhibiting SRC kinase activity during wound healing (By similarity).
- Localized to the cytoplasm, KRT6A is expressed in the corneal epithelium and is associated with ectodermal dysplasia and palmoplantar keratoderma when mutated; it is also recognized as an allergen.

- The immunogen is a synthetic peptide from the N-terminal region (amino acids 1–100) of human KRT6A; this design may provide specificity for KRT6A among highly homologous type II keratins.
- For Western blotting, a predominant band at approximately 60 kDa is expected; validate in the relevant sample system and consider loading controls for normalization.
- In immunohistochemistry (paraffin), antigen retrieval conditions (pH, heat, time) should be optimized for tissue type; confirm staining specificity with appropriate controls.
- The antibody also detects mouse and rat KRT6 proteins, supporting translational studies; verify cross-reactivity in your experimental models by positive/negative controls.
