Biological Background: Cytokeratin 5 Function and Localization
- Keratin, type II cytoskeletal 5 (KRT5), also designated Cytokeratin-5 (CK-5) or 58 kDa cytokeratin, is a type II keratin encoded by the KRT5 gene in humans.
- As a structural component of intermediate filaments, KRT5 partners with its obligate type I keratin 14 (KRT14) to form a resilient cytoskeletal network that provides mechanical integrity to basal keratinocytes of stratified epithelia. Related references: PMID:1372711, PMID:1694855
- KRT5 modulates immune cell recruitment by regulating the abundance of macrophage chemotactic cytokines and CTNND1 localization in keratinocytes, thereby influencing Langerhans cell homing to the epidermis.
- This keratin localizes to the cytoplasm, where it assembles into intermediate filaments.
- Expression is predominantly observed in keratinocytes and the corneal epithelium, as confirmed by protein-level detection.
- KRT5 is a phosphoprotein with a coiled-coil domain structure, and genetic variants are linked to epidermolysis bullosa simplex.

- The immunogen corresponds to the N-terminal region (amino acids 1-100) of human KRT5, which may recognize full-length protein; validate in the relevant sample system.
- For Western blot, the predicted molecular weight is approximately 62 kDa; note that phosphorylation or other modifications may cause migration shifts.
- Suitable for immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC); consider optimization of antigen retrieval and antibody dilution for each tissue type.
- Cross-reactivity with mouse allows for rodent model studies; however, sequence alignment is advised to confirm epitope conservation.
