Biological Background: Cytokeratin 2e Function and Localization
- Keratin, type II cytoskeletal 2 epidermal (KRT2), also known as cytokeratin-2e (CK-2e), is a type II keratin encoded by the KRT2 gene, with a molecular weight of approximately 65 kDa.
- KRT2 is a structural component of intermediate filaments in suprabasal keratinocytes of the stratified epidermis, providing mechanical integrity and resilience to the differentiating epidermis (PubMed:12598329, PubMed:1380918). Related references: PMID:12598329 PMID:1380918
- It forms heteropolymers with type I keratins, such as KRT10, assembling into keratin intermediate filament networks essential for epidermal differentiation (PubMed:12598329). Related references: PMID:12598329
- Contributes to terminal differentiation and cornification of keratinocytes, playing a role in establishing the epidermal barrier (PubMed:9804344). Related references: PMID:9804344
- Expressed specifically in the upper spinous and granular suprabasal layers of normal adult epidermis from most body sites, but absent in foreskin and certain pathological tissues.
- Localizes to the cytoplasm, consistent with its role as an intermediate filament protein.
- Post-translational modifications include methylation and phosphorylation; associated with the disease ichthyosis.
- Keywords: Coiled coil, Cytoplasm, Disease variant, Ichthyosis, Intermediate filament, Keratin, Methylation, Phosphoprotein.
Experimental Guidance and Technical Tips
- The immunogen used for this antibody is a recombinant fusion protein covering amino acids 293–422 of human KRT2, a region within the rod domain; consider epitope specificity in your experimental design.
- This antibody is validated for WB, IF-P, IHC-P, and ELISA. For WB, the predicted band is around 65 kDa; validate in relevant tissue or cell lysates expressing KRT2.
- For immunohistochemistry, antigen retrieval and titration should be optimized in your sample system.
- Note that KRT2 expression is restricted to suprabasal layers; positive control tissues should include skin from body sites such as thigh or foot sole.
