Biological Background: Cytokeratin 20 (KRT20) Function and Localization
- Keratin, type I cytoskeletal 20 (KRT20), also known as Cytokeratin-20 (CK-20), Keratin-20 (K20), or Protein IT, is a type I keratin intermediate filament protein.
- It plays a significant role in maintaining keratin filament organization in intestinal epithelia. When phosphorylated, KRT20 is involved in the secretion of mucin in the small intestine (By similarity).
- Subcellular localization is cytoplasmic, consistent with its role as a cytoskeletal component.
- Predominantly expressed in intestinal epithelium, with specific localization in luminal cells of colonic mucosa. Additionally expressed in Merkel cells of keratinized oral mucosa, including gingival rete ridges, palatal basal layer, and lingual taste buds.
- Classified under UniProt keywords: Apoptosis, Coiled coil, Cytoplasm, Direct protein sequencing, Intermediate filament, Keratin, Phosphoprotein, Proteomics identification, Reference proteome.
- The protein has a molecular weight of approximately 48,487 Da (UniProt) but migrates at ~48 kDa in SDS-PAGE.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central portion of human KRT20 (aa 245-424), which is conserved in mouse; cross-reactivity with mouse has been validated. Consider sequence alignment for other species before use.
- For Western blotting, a band at ~48 kDa is expected. Optimize blocking and antibody dilution using a relevant cell lysate (e.g., Caco-2 or HT-29 intestinal cells).
- In immunofluorescence/immunocytochemistry, test fixation and permeabilization conditions; validate nuclear vs. cytoplasmic staining in intestinal cell lines.
- For ELISA, use recombinant KRT20 or a specific peptide as coating antigen; perform a titration to determine optimal working concentration.
- As a polyclonal antibody, lot-to-lot consistency should be verified in the intended application.
