  • Cytokeratin 17, also known as keratin 17 (K17), is a type I intermediate filament protein and a member of the keratin family, characterized by a coiled-coil central rod domain.
  • It plays a critical role in the formation and maintenance of skin appendages, particularly in determining hair shape and orientation, and is required for the persistence of the anagen (growth) phase of the hair follicle.
  • K17 modulates the function of tumor necrosis factor-alpha (TNF-α) specifically in the context of hair cycling, influencing inflammatory and proliferative responses.
  • The protein regulates epithelial cell growth and protein synthesis by binding to the adapter protein stratifin (SFN) and activating the Akt/mTOR signaling pathway.
  • As a promoter of epithelial proliferation, K17 shapes the immune microenvironment by favoring a Th1/Th17-dominated response, thereby contributing to the development of basaloid skin tumors.

  • The immunogen for this antibody is a synthetic peptide spanning amino acids 333–432 of human K17. This region lies near the C-terminal tail domain, which may be less conserved across keratins; therefore, cross-reactivity with other keratin family members should be assessed.
  • For Western blot, a band at approximately 48 kDa is expected; reducing conditions and adequate denaturation are recommended as K17 is a filament-forming protein.
  • In immunofluorescence and IHC, permeabilization and antigen retrieval steps may be necessary to expose the cytoplasmic intermediate filament epitope. Validate using positive control tissues such as skin, hair follicle, or epithelial cell lines known to express K17.
  • The antibody cross-reacts with mouse and rat, making it suitable for studies in common model organisms. However, verify reactivity in your specific assay conditions due to potential species-specific epitope variations.
