Biological Background: Cytokeratin 10 Function and Localization
- Keratin 10 (CK-10, K10) is a type I intermediate filament protein encoded by the KRT10 gene.
- In suprabasal keratinocytes, CK10 forms heteropolymers primarily with the type II keratin KRT1, contributing to the intermediate filament network essential for epidermal differentiation, mechanical integrity, and barrier function.
- Belongs to the keratin family, characterized by a central α-helical rod domain flanked by non-helical head and tail domains; post-translationally modified by phosphorylation and disulfide bond formation.
- While primarily cytoplasmic as part of the cytoskeleton, CK10 is also detected in the extracellular space and on the cell surface, particularly in differentiated epithelial cells.
- Expressed in all suprabasal layers of stratified epithelia, including the stratum corneum. Additionally, CK10 is found on the surface of desquamated nasal epithelial cells and lung cell lines. Related references: PMID:12427098 PMID:19627498
- CK10 serves as a receptor for Staphylococcus aureus on desquamated nasal epithelial cells, interacting with clfB and thereby promoting nasal colonization. Related references: PMID:12427098
- CK10 binds Streptococcus pneumoniae PsrP on lung cell lines, facilitating bacterial adherence; this interaction does not require glycosylation of either protein. Related references: PMID:19627498
Experimental Guidance and Technical Tips
- Immunogen design: The antibody was raised against a synthetic peptide spanning the C-terminal region (amino acids 485–584) of human CK10, which is predicted to be specific to the CK10 sequence and less conserved among other keratins.
- Application suitability: This antibody is suitable for immunofluorescence on fixed/permeabilized samples (IF-P) and ELISA. For IF-P, optimize fixation and permeabilization to preserve intermediate filament structure; for ELISA, titrate antibody and validate in the relevant sample matrix.
- Cross-reactivity: The antibody shows reactivity with human, mouse, and rat CK10, making it useful for studies in rodent models.
