Biological Background: Cytokeratin 1 (KRT1) Function and Localization
- Keratin, type II cytoskeletal 1 (KRT1), also designated cytokeratin-1 (CK-1) or 67 kDa cytokeratin, is a type II intermediate filament protein encoded by the KRT1 gene in humans.
- As a structural component of intermediate filaments, KRT1 forms heteropolymers with type I keratins, providing mechanical strength and stability to differentiating suprabasal keratinocytes of stratified epithelia. Related references: PMID:1381288
- Through interactions with integrin beta-1 (ITB1) and receptor of activated protein C kinase 1 (RACK1), KRT1 may regulate the activity of kinases such as PKC and SRC. Related references: PMID:17956333
- In complex with C1QBP, KRT1 serves as a high-affinity receptor for kininogen-1 (HMWK), linking intermediate filaments to cell signaling. Related references: PMID:21544310
- Subcellularly, KRT1 localizes to both the cytoplasm and the cell membrane, consistent with its structural and receptor functions.
- KRT1 is predominantly expressed in terminally differentiating epidermis; the protein source for characterization is often neonatal foreskin.
- This keratin undergoes several post-translational modifications, including phosphorylation, methylation, and citrullination, and is associated with skin disorders such as ichthyosis and palmoplantar keratoderma when mutated.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 545–644 in the C-terminal region of human KRT1. This region is likely surface‑exposed in assembled filaments, making the antibody suitable for detection of both denatured and native KRT1 under appropriate conditions.
- For Western blotting (WB), the predicted molecular weight of KRT1 is approximately 66 kDa. Include proper reducing agents and denaturing steps; validate detection in your specific cell or tissue lysates, as keratin extraction may require high‑salt or urea buffers.
- In immunofluorescence of paraffin‑embedded sections (IF‑P), antigen retrieval is essential. A permeabilization step may improve accessibility of the intracellular target. Confirm reactivity with human, mouse, or rat skin specimens.
- For ELISA, pair this monoclonal antibody with an appropriate type I keratin detection antibody if developing a sandwich immunoassay. Titrate both capture and detection reagents in your assay system.
- As cross‑reactivity is noted for human, mouse, and rat, consider sequence alignment of the immunogen region to verify epitope conservation and employ species‑specific controls.
