Biological Background: CYP51A1 (Lanosterol 14-alpha demethylase) Function and Localization
- CYP51A1 encodes lanosterol 14-alpha demethylase, a cytochrome P450 monooxygenase, also known as LDM, CP51, CYP51, CYPL1, P450L1, and P450-14DM.
- It catalyzes the three-step oxidative removal of the 14α-methyl group from sterol intermediates such as lanosterol and 24,25-dihydrolanosterol, a critical step in the cholesterol biosynthesis pathway. Related references: PMID:20149798, PMID:8619637, PMID:9559662
- The enzyme localizes to the endoplasmic reticulum membrane and microsomal fraction, consistent with its role in sterol metabolism.
- Ubiquitously expressed with highest levels in testis, ovary, adrenal, prostate, liver, kidney, and lung, reflecting widespread requirement for cholesterol synthesis.
- Contains a transmembrane helix, heme-binding domain, and iron cofactor essential for its monooxygenase activity, classifying it as an oxidoreductase.
- Alternatively spliced transcripts have been reported, and the protein may undergo ubiquitin-like (Ubl) conjugation, indicating regulatory diversity.
- The predicted molecular weight is approximately 57.3 kDa, aligning with the observed 57 kDa band in Western blot applications.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 210-509, covering a conserved region; cross-reactivity with mouse and rat is provided, but validation in target species is recommended.
- For Western blot, expect a band near 57 kDa; optimal loading controls and tissue lysates with high CYP51A1 expression (e.g., testis, liver) should be considered.
- ELISA protocols may require optimization of coating antigen (recombinant protein or cell lysates) and antibody dilution to achieve optimal signal-to-noise ratio.
- Due to the ubiquitous but tissue-variable expression, include both positive and negative control samples to confirm specificity.
- Shipping on ice bag ensures antibody stability; upon receipt, aliquot and store at the appropriate temperature as per standard antibody handling practices.
